Virtually every competent drug discovery organization has criteria for the transition between hit to lead and lead optimization. Virtually nobody publishes these criteria because the organizations consider them proprietary. By virtue of very fast note taking and a combined effort, a Pfizer colleague and I were able to capture the hit to lead exit criteria from an oral presentation by a medicinal chemist from the AZ organization. Operator independence. The GEX method is virtually operator independent. The only inputs required before fitting are parameters concerning the precision of the numerical integration and exit criteria for the Marquardt algorithm. The same set of Inputs was used for all five MWDs. Currently the method also requires upper and lower limits on the GEX parameters but a simple modification of the code can eliminate this need. The life-cycle approach has attracted broad acceptance across the pharmaceutical and healthcare industries and can be refined to meet the needs of particular applications. Different organizations use variants of the life cycle, but the methodology of dividing a life cycle into phases remains the same. Two doses of oxcarbazepine have been compared in a double-blind, parallel-group, randomized trial in patients with uncontrolled partial-onset epilepsy who had previously taken carbamazepine monotherapy (10). After two open phases in 143 patients, 96 were randomized to oxcarbazepine 300 or 2400 mg/day for 126 days. The time to meet an exit criterion was significantly in favor of oxcarbazepine 2400 mg/day. In aU, 24 of the 47 non-rando-mized patients withdrew because of an adverse event, most commonly dizziness, ataxia, headache, nansea, vomiting, or fatigne. Three withdrew becanse of laboratory abnormalities, one each with leukopenia, hjrponatre-mia, and hjrperglycemia. Headache, dizziness, and nansea were the only adverse events that occurred in more than 10% in either gronp. If a molecule is strained, atoms may not be ver y close to the minimum of their individual potential energy wells when the best compromise geometry is reached. In this study, the relationship between oscillatory heat release and large vortex structure was systematically examined as a function of flow and chemistry scaling. Figure 16.3 shows the experimental setup used for producing vortex flames. A premixed propane-air jet was ignited, and the flame was stabilized at the exit downstream of a sudden-expansion flameholder. A 75-watt compression driver was used to apply controlled disturbance and to produce periodic vortices into a jet flame. The frequency response of actuation was evaluated separately to maintain a similar amplitude of acoustic disturbance at the exit plane. Brutin et al. [9] studied the transition from steady to unsteady flow boiling and developed a transition criterion. This study was performed experimentally in minichannels but the theory also applies to confined geometries such as microchannels. This criterion is based on observations of the unsteady two-phase flow the two-phase flow stops at a location in the microchannel, then all the fluid after this location is expelled toward the exit, whereas all the fluid before this location is sent back to the entrance.
