After electroporation into Raji cells, CD4 antigen-positive cells are enriched by repeated cell sorting and plasmids are recovered in Escherichia coli. In this study, we report comparisons of the activities and thermal stabilities of purified and recombinant P. funiculosum and T. reesei Cel7A enzymes. All assays were conducted in the presence of the catalytic domain of A. cellulolyticus El endoglucanase (Elcd) as expressed in Escherichia coli and used the dialysis saccharification assay (DSA) format reported by Baker and coworkers . We also report the cloning of the cel7A gene from P. funiculosum in E, coli and exploration of several alternative signal peptide configurations.
