Dulcolax - Bisacodyl

It should be noted that routine crushing of tablets should not be recommended. The bioavailability of marketed tablets should be satisfactory. Moreover, crushing of some tablets (e.g. Dulcolax [bisacodyl]) can cause severe side effects, while crushing of tablets intended for controlled or sustained release can cause dose dumping and dangerously high blood concentrations of drug. [Pg.174]

Bisa.codyl, 4,4 -(2-PyridyLmethylene)bisphenol diacetate [603-50-9] (Dulcolax) (9) is a white to off-white crystalline powder ia which particles of 50 p.m dia predominate. It is very soluble ia water, freely soluble ia chloroform and alcohol, soluble ia methanol and ben2ene, and slightly soluble ia diethyl ether. Bisacodyl may be prepared from 2-pyridine-carboxaldehyde by condensation with phenol and the aid of a dehydrant such as sulfuric acid. The resulting 4,4 -(pyridyLmethylene)diphenol is esterified by treatment with acetic anhydride and anhydrous sodium acetate. Crystallisation is from ethanol. [Pg.201]

Bisacodyl is a laxative that is indicated in short-term treatment of constipation evacuation of colon for rectal and bowel evaluation preparation for delivery or surgery. Bisacodyl is the only diphenyhnethane derivative available in the United States. It is marketed as an enteric-coated preparation (Dulcolax, Correctol, others) and as a suppository for rectal administration. The usual oral daily dose of bisacodyl is 10 to 15 mg for adults and 5 to 10 mg for children 6 to 12 years old. The drug requires hydrolysis by endogenous esterases in the bowel for activation, and so the laxative effects after an oral dose usually are not produced in less than 6 hours taken at bedtime, it will produce its effect the next morning. Suppositories work much more rapidly. [Pg.108]
