Dosage form Pulmozyme Inhalation Solution is a sterile, preservative-free, highly purihed solution of recombinant human deoxyribonuclease I (dornase alfa). Each Pulmozyme single-use ampule will deliver 2.5 ml of the solution to the nebulizer bowl. The aqueous solution contains domase alfa l.Omg/ml. 

F. Role in therapy Pulmozyme is a mucolytic enzyme used in the treatment of cystic hbrosis. Although it is not a cure, domase alfa is an effective mucolytic for adjunctive treatment. AU compliant patients with cystic fibrosis, irrespective of disease severity, who produce purulent sputum are potential candidates for Pulmozyme therapy. It is useful for liquefying the thick mucus secreted by cystic fibrosis patients, and causes both objective improvement (as measured by pulmonary function testing) and subjective symptomatic improvement. Pulmozyme reduces the frequency of respiratory infections requiring parenteral antibiotics and improves pulmonary function.

Twenty-seven out of 44 FDA-approved biopharmaceuticals have been tested in a battery of genotoxicity assays. Eighty-five different assays performed yielded negative results. The most commonly performed assays were the Ames test, the chromosomal aberration assay in human lymphocytes, the mouse lymphoma gene mutation assay, and the mammalian in vivo erythrocyte micronucleus test. Examples of the range of biopharmaceutical products tested include, domase alfa (deoxyribonuclease I-DNAse), trastuzumab (mAb to human epidermal growth factor receptor 2), alteplase (tissue plasminogen activator), infliximab (mAb to the human tumor necrosis factor a).

Domase alfa is a mncolytic agent that cleaves DNA released by neutrophils that are mobilized to respiratory tract in response to infection, redncing viscoelasticity of pnrnlent Inng secretions, increasing airflow, and decreasing risk of infection. It is nsed in the treatment of cystic fibrosis.
