Characteristic properties of endopectate lyases are the high pH optimum, and a requirement for Ca2+ ions in order to maintain catalytic activity. The pH optimum of various endopectate lyases ranges from 8.0 to 9.5 (Refs. 4, 178, 234, 236, 243). Besides activation by Ca2+ ions, the optimal concentration of which is 1 mM,234,236,244 strontium salts were also considered in the case of Bacillus sp. lyase.234 The enzyme from Pseudomonas sp. was also partly activated by magnesium chloride,178 and for the lyase of Clostridium felsineum, salts of other bivalent cations had an activating effect as well.245 (Ethylenedinitrilo)tetraacetic acid completely inactivated all of the lyases mentioned. The activity of endopectate lyase from Pseudomonas was also lessened in the presence of sodium chloride, potassium chloride, and dipotassium hydrogen phosphate (K2HP04).

Cabon tetrachloride, n-hexane, chloroform, ACN, acetone, THF, pyridine, acetic acid, and their various mixtures were applied as mobile phases for adsorption TLC. Methanol, 1-propanol, ACN, acetone, THF, pyridine and dioxane served as organic modifiers for RP-TLC. Screening of ENRO, CIPRO, DAN, NOR, FLU, OXO, and NALA in pork muscle was based on HPTLC after the SPE procedure (195). Tissue samples were extracted with MeCN-NaOH, the supernatant dried, and the residues dissolved in dipotassium hydrogen phosphate buffer (pH 7.4) hexane was added. The aqueous phase was cleaned up using the SPE technique. After washing the cartridge, analytes were eluted with MeOH-ammonia solution (75 25). After the preparation step, extracts were spotted and eluted on silica gel plates. The plate is first inspected under UV illumination at 312 nm and then sprayed with terbium chloride. P-E-M Buffer Dissolve 8.8 g of potassium dihydrogen phosphate (KH2PO4) and 8.0 g of dipotassium hydrogen phosphate trihydrate (K2HP04-3H20) in about 900 mL of water. Potassium pyrophosphate, K4P207.—The pyrophosphate is produced by heating dipotassium hydrogen phosphate, or by neutralizing anhydrous phosphoric acid with potassium hydroxide dissolved in absolute alcohol. Its specific heat is 0-1910 between 17° and 98° C.11 It is soluble in water, forming a slightly alkaline solution, and yields a trihydrate. 

Only telrapotassium diphosphate is of major industrial importance, due to its high water solubility. It is produced in an analogous way to the sodium compound from dipotassium hydrogen phosphate.

The composition of the fungal medium used for the transformation studies was, sucrose(1.50 gm), glucose(1.50 gm), polypeptone(0.50 gm), dipotassium hydrogen phosphate(0.10 gm), magnesium sulfate(Q.50 gm), potassium chloride(0.50 gm) and ferrous sulfate heptahydrate(0.001 gm) in 100 ml of distilled water.
