Danazol Hormonal contraceptives

The synthesis of Lp(a) can be influenced by hormones, as is reported by Albers et al. (A7), who observed a reduction of Lp(a) concentrations in nor-molipidemic postmenopausal women treated during 6 weeks with the anabolic steroid stanazolol. The same effect was reported for norethisterone (FI) and danazol (Cl2). Exogenous estrogens, as used in treatment of postmenopausal complaints and in oral contraceptives, induce a decrease of Lp(a) (D9, H27, K13, KAO, L21, S37). This effect is dose-related and dependent on Lp(a) levels before treatment (F2). Watanabe (W9) investigated the influence of progestogen (alone) and detected a decrease of Lp(a) during treatment. 

Sex hormones and hormone antagonists. Oestrogens increase the synthesis of some vitamin K dependent clotting factors and progestogen-only contraceptives are preferred. The hormone antagonists danazol, flutamide and tamoxifen enhance the effect of warfarin. 

The most common drugs responsible for acneform eruptions are ACTH, androgenic hormones, anticonvulsants (hydantoin derivatives, phenobarbital, trimethadione), corticosteroids, danazol, disulfiram, halogens (bromides, chlorides, iodides), lithium, oral contraceptives, tuberculostatics (ethionamide, isoniazid, rifampin), vitamins B2, B6, and B12. 

Ovulation suppression (e.g., oral contraceptives, danazol, gonadotropin-releasing hormone agonists) should be used before resorting to surgical therapy in women with severe PMDD.

All medical therapies (nonsteroidal anti-inflammatory drugs, oral contraceptives, progestins, danazol, or gonadotropinreleasing hormone agonists) are equally efficacious in treating endometriosis-related pain based on available evidence. Choice among agents is determined primarily by side-effect profile, cost, and individual patient response. 

There is a theoretical risk that the effects of danazol or gestrinone and hormonal contraceptives might be altered or reduced by concurrent use.
