Columns, packed plate

Hsieh and Jorgenson prepared 12-33- 4m HPFC columns packed with 5.44-pm spherical stationary phase particles. To evaluate these columns they measured reduced plate height, h,... [Pg.615]

Packed Columns versus Plate Columns Packed columns are usually specified when plate devices would not be feasible because of undesirable fluid charac teristics or some special design requirement. Conditions favoring packed columns are ... [Pg.1385]

After column packing, each column is tested for theoretical plate count, peak symmetry, resolution, pore volume, and back pressure. If one of these tests fails the column is removed from the production cycle. If a PSS SEC column is kept in storage for a longer time, it is retested for theoretical plate count, peak symmetry, resolution, pore volume, and back pressure prior to shipping to the customer to prove up-to-date column performance. [Pg.289]

Each SynChropak column is tested chromatographically to assure that it has been packed according to specifications. For SynChropak GPC columns, a mixture of a high molecular weight DNA and glycyltyrosine, a dipeptide, is used to evaluate internal volume and efficiency. The mobile phase used for the test is 0.1 M potassium phosphate, pH 7, and the flow rate is 0.5 ml/min for 4.6-mm i.d. columns. Minimum plate count values and operational flow rates are listed in Table 10.4 for 4.6-mm i.d. columns of all supports and the various diameters of the SynChropak GPC 100 columns. [Pg.314]

Direct contact heal exchangers should be considered whenever the process stream and the coolant are compatible. The equipment used is basically simple and cheap and is suitable for use with heavily fouling fluids. For liquids containing solids, spray chambers, spray columns, and plate and packed columns are used. [Pg.137]

Figure 1.17 Separation of large ring polycyclic aroaatic hydrocarbons extracted from carbon black on a 1.8 x 0.2 n I.D. fused silica capillary column packed with 3 micrometer spherical octadecylsllanized silica gel eluted with a stepwise solvent gradient at a flow rate of 1.1 mlcroliters/min with an inlet pressure of about 360 atmospheres. Under isocratic conditions this column yielded ca. 225,000 theoretical plates. (Reproduced with permission from ref. 238. Copyright Friedr. Vieweg t Sohn). Having chosen the test mixture and mobile diase composition, the chromatogram is run, usually at a fairly fast chart speed to reduce errors associated with the measurement of peak widths, etc.. Figure 4.10. The parameters calculated from the chromatogram are the retention volume and capacity factor of each component, the plate count for the unretained peak and at least one of the retained peaks, the peak asymmetry factor for each component, and the separation factor for at least one pair of solutes. The pressure drop for the column at the optimum test flow rate should also be noted. This data is then used to determine two types of performance criteria. These are kinetic parameters, which indicate how well the column is physically packed, and thermodynamic parameters, which indicate whether the column packing material meets the manufacturer s specifications. Examples of such thermodynamic parameters are whether the percentage oi bonded... [Pg.184]

A particular column can be used for different types of LC by changing the eluent components. For example, a column packed with RP-18 bonded silica gel can be used for SEC with THF, NPLC with n-hexane, and RPLC with aqueous acetonitrile. When separation cannot be achieved by improving the theoretical plate number of a column, it may be achieved by selection of an appropriate stationary phase material and/or eluent. [Pg.231]
