Ciclopirox

Ciclopirox may be produced as follows 2 g of 4-methyl-6-cyclohexyl-2-pyrone were heated with 1 g of hydroxylamine hydrochloride and 5 g of 2-aminopyridine to B0°C for B hours. [Pg.339]

Reaction of ciclopirox with ethanolamine gives the desired product. [Pg.340]

Onychomycosis is a chronic infection that rarely remits spontaneously. Adequate treatment is essential to prevent spread to other sites, secondary bacterial infections, cellulitis, or gangrene. Due to the chronic nature and impenetrability of nails, topical agents have low efficacy rates for treating onychomycosis. Oral agents that can penetrate the nail matrix and nail base, such as itraconazole and terbinafine, are more effective than ciclopirox lacquer. Itraconazole and terbinafine demonstrate mycological cure rates of 62%37 and 76%,38 respectively, while ciclopirox has a cure rate of 29% to 36%.39... [Pg.1207]

Ciclopirox olamine is a hydroxypyridone antifungal that is structurally unrelated to other antifungal agents. It is a broad-spectrum antimycotic agent with inhibitory activity against dermatophytes and yeasts such as Candida species and Pityrosporum orbicu-lare. Although some absorption may occur adverse reactions are rare. Local pruritus may occur. [Pg.481]

Ciclopirox olamine (Loprox) is a pyridone derivative available for the treatment of cutaneous dermatophyte infections, cutaneous C. albicans infections, and tinea versicolor caused by Malassezia furfur. It interferes with fungal growth by inhibiting macromolecule synthesis. [Pg.602]

Ciclopirox olamine is a synthetic broad-spectrum antimycotic agent with inhibitory activity against dermatophytes, Candida species, and P orbiculare. This agent appears to inhibit the uptake of precursors of macromolecular synthesis the site of action is probably the fungal cell membrane. [Pg.1289]

Pharmacokinetic studies indicate that 1-2% of the dose is absorbed when applied as a solution on the back under an occlusive dressing. Ciclopirox olamine is available as a 1% cream and lotion (Loprox) for the topical treatment of dermatomycosis, candidiasis, and tinea versicolor. The incidence of adverse reactions has been low. Pruritus and worsening of clinical disease have been reported. The potential for delayed allergic contact hypersensitivity appears small. [Pg.1289]

Topical 8% ciclopirox olamine (Penlac nail lacquer) has been approved for the treatment of mild to moderate onychomycosis of fingernails and toenails. Although well tolerated with minimal side effects, the overall cure rates in clinical trials are less than 12%. [Pg.1289]

Each gram of PENLAC NAIL LACQUER (ciclopirox) topical solution, 8%, contains 80 mg ciclopirox in a solution base consisting of ethyl acetate, isopropyl alcohol,... [Pg.103]
