Nitrogen sources include proteins, such as casein, zein, lactalbumin protein hydrolyzates such proteoses, peptones, peptides, and commercially available materials, such as N-Z Amine which is understood to be a casein hydrolyzate also corn steep liquor, soybean meal, gluten, cottonseed meal, fish meal, meat extracts, stick liquor, liver cake, yeast extracts and distillers solubles amino acids, urea, ammonium and nitrate salts. Such inorganic elements as sodium, potassium, calcium and magnesium and chlorides, sulfates, phosphates and combinations of these anions and cations in the form of mineral salts may be advantageously used in the fermentation. [Pg.1062]

Differential Solubility Methods. Numerous methods have been developed to obtain one or more of the various caseins from whole casein or directly from skim milk based on their differential solubility (Thompson 1971 Mackinlay and Wake 1971 Whitney 1977). While some early procedures indicated the possibility of fractionating whole casein into different components, it was not until the 1950s that systematic procedures were proposed for the fractionation of casein into Warner s a-, 0-, and y-caseins. Hipp et al. (1952) developed two procedures which have been used extensively or partially incorporated into other methods. The first is based upon the differential solubilities of the caseins in 50% alcohol in the presence of ammonium acetate by varying the pH, temperature, and ionic strength. In addition, to release lipids from source material, such as those in starch, fish meal, or milk, it might be necessary to treat the sample with an acid prior to lipid extraction (see Basic Protocol 4). In the case of milk, addition of ammonium hydroxide is necessary to dissolve casein prior to lipid extraction, which will release the lipids from its surrounding matrix (e.g., from the film surrounding the fat globules in milk). Furthermore, in certain cases, it is necessary to predry the sample in order to allow efficient and complete extraction of lipids. Casein which is a phosphoprotein, is the chief nitrogenous constituent of milk, in which it occurs as a calcium salt in combination with calcium phosphate. Casein is insoluble in water, but its salts are readily soluble those of calcium, barium, strontium, and magnesium form opalescent colloidal solutions.
