A single measurement of a calibration sample can give the concentration of the test solution by a simple ratio. This is often done in techniques where a calibration internal standard can be measured simultaneously (within one spectrum or chromatogram) with the analyte and the system is sufficiently well behaved for the proportionality to be maintained. Three primary methods are used for calibration external standard calibration, internal standard calibration, and standard addition. Figs. 1-3 show the typical response curves for each method. 

Quantitative analysis using external calibration, internal standard calibration, and the method of standard additions are all possible with SPME. Calibration is discussed in Section 1.5.2 and at greater length in Chapter 2. 

Infrared spectra were obtained on a Digilab FTS-15/B spectrometer. Four hundred scans at a resolution of 2 cm l were signal averaged and the spectra were stored on a magnetic disc system. The frequency scale was calibrated internally with a helium-neon laser to an accuracy of 0.2 cm l. Spectra recorded at elevated temperatures were obtained using a SPECAC high temperature cell and controller mounted in the spectrometer. 

Quantification methods can be based on standard additions or external calibration. Internal standards may be used to correct for some losses of material but for many analyses suitable compounds are unavailable. In LC-MS and LC-MS/MS the ideal internal standards are stable isotope analogs of the target compounds as these behave exactly as the native compound used but can be distinguished by their different masses. For LSD-related compounds with slightly different structures such as methysergide have been used, but trideuterated LSD is available commercially. 

The graph in Fig. 1 shows a separating efficiency measurement, in which the number of theoretical stages per metre is entered over the vapour load in the form of the F-factor. When the column packings are used with actual material systems, reference can be made to this calibration. Miniplant columns with calibrated internal fittings then make it possible to increase the scale of the equipment directly from the miniplant scale to the production scale. This offers the advantage of a fast and cost-effective process development.

As with external calibration, internal calibration relies on the plotting of a response function over a given concentration range, then using the response function to establish the concentrations of the dosed samples in analytes. The fundamental difference between the two dosage methods is based on the fact that in internal calibration, a molecule (standard) is added systematically at a known concentration to the solution to be analyzed.
