Tromethamine buffer

Another option for patients with severe acidemia is tromethamine (THAM). This inert amino alcohol buffers acids and C02 through its amine (—NH2) moiety ... [Pg.427]

Mann and Mitchell [58] described a simple colorimetric method for estimation of (-D)-penicillamine in plasma. Blood containing 2-50 pg of penicillamine was mixed with 0.1 M EDTA solution in tromethamine buffer solution. 0.1 mL of this solution was adjusted to pH 7.4 and centrifuged. To a portion of the plasma was added 3 M HCL, the mixture was freeze-dried, and a suspension of the residue in ethanol was centrifuged. The supernatant liquid was mixed with tromethamine buffer solution (pH 8.2) and 10 mM 5,5 -dithiobis-(2-nitrobenzoic acid) in phosphate buffer solution (pH 7.0), the mixture was shaken with ethyl ether, and the absorbance of the separated aqueous layer was measured at 412 nm. The mean recovery was 60% (four determinations), and the calibration graph was linear for the cited range. [Pg.145]

Following rinsing in PBS, the sections are treated with free biotin solution (2.5 mg in 0.1 mM PBS) for 4 min to saturate the remaining binding sites of the egg white avidin. The sections are thoroughly rinsed with tromethamine-based buffer or PBS, followed by the following sequential immunostaining protocol biotinylated secondary antibody,... [Pg.100]

THAM, available as a 0.3 N solution, is a highly alkaline, sodium-free organic amine that acts as a proton acceptor to prevent or correct acidosis. Tromethamine combines with hydrogen ions from carbonic acid to form bicarbonate and a cationic buffer. THAM also acts as an osmotic diuretic to increase urine flow, urine pH, and the excretion of fixed acids, CO2, and electrolytes. At pH 7.4, 30% of THAM is not ionized and therefore may penetrate into cells and neutralize acidic anions of the intracellular fluid. Intracellular pH increases have been noted within 1 hour after the infusion of THAM. There is, however, no chnical or physiologic evidence that this action is beneficial, or that THAM is more efficacious than sodium bicarbonate. " ... [Pg.992]

The pKa of I (azathioprine) was determined by solubility and spectrophotometric methods. Triplicate samples of I in masses that were incompletely soluble were equilibrated at 25 C for 48 h with 10 mL of 0.(EN buffer at pH 4.00 and with 10 mL of 0.025M tromethamine buffers at six pH values from 7.00 to 8.60. The pH values of samples at 25 were recorded, th die filtrates were diluted and I concentraticms were determined from absorbances at 280 nm comparison to known I solutions at the same pH values. [Pg.86]
