Selenium is required, but levels must fall into a narrow window. Both deficiency and toxicity symptoms occur. The element is also used therapeutically in cancer treatment. It is the co-factor of the enzyme glutathione peroxidase which is thought to play an important role in oxygen toxicity. The determination of Se in blood or serum is not easy, as many incorrect, inaccurate and imprecise methods have been published (Magee and James 1994). A suggested procedure for Se in body fluids is based on GF-AAS (Thomassen et al. 1994)- For tissues SS-AAS may be used (Fler-ber 1994a). Recent developments by Turner et al. (1999) show that LC-ICP-MS is sensitive and reproducible at low levels. [Pg.203]

Isolated liver perfusion systems are commercially available (e.g.. Harvard Apparatus, Holliston, MA). Commonly used perfusates contain various amounts of albumin and red blood cells (RBCs) that affect the protein binding of the compound as well as other components that may affect the metabolism of the test compound and hence the bioavailability of the eompoimd. Heparinized whole blood from the same species is perhaps the best perfusate. However, heparinized whole blood is not always readily available and potential issues such as the formation of clots need to be taken into consideration. Bovine blood, bovine serum albumin, and bovine or human erythrocytes have also been used to mimic in vivo conditions because they provide hemoglobin as the oxygen carrier and ideal protein binding conditions. [Pg.169]

There is a paucity of clinical trial evidence comparing the benefit of diuretics to other therapies for symptom relief or long-term outcomes. Additionally, excessive preload reduction can lead to a decrease in CO resulting in reflex increase in sympathetic activation, renin release, and the expected consequences of vasoconstriction, tachycardia, and increased myocardial oxygen demand. Careful use of diuretics is recommended to avoid overdiuresis. Monitor serum electrolytes such as potassium, sodium, and magnesium frequently to identify and correct imbalances. The level of EPO production in the kidneys (or liver) is primarily regulated by the oxygen demand of the producer cells, relative to their oxygen supply. Under normal conditions, when the producer cells are supplied with adequate oxygen via the blood, EPO (or EPO mRNA) levels are barely detectable. However, the onset of hypoxia (a deficiency of oxygen in the tissues) results in a very rapid increase of EPO mRNA in producer cells. This is followed within 2 h by an increase in serum EPO levels.
