Since the first successful transformation of tobacco chloroplasts [83], expression systems based on the transformation of plant plastids has attracted the attention of plant biotechnologists. The features that make this technology so attractive are its potential for high protein yield, along with inherent biosafety features such as limited plastid transfer via pollen (due to maternal inheritance of plastid-encoded genes) [84] and the relatively low probability of transgene movement from the chloroplast to the nucleus [85, 86]. 

The main biosafety features of the fermentation system include the use of steam barriers on double O-ring seals, supply lines and mechanical seals on stirrer shafts, multiple O-ring seals, piping of condensate lines and pressure relief systems to a kill-tank , double (in series) filtration of inlet... 

The "P" has been replaced with "BSL" or Biosafety Level. There are four biosafety levels which are defined according to a combination of facility design, laboratory practices and techniques, equipment and health and safety controls. It is not practical to try to completely describe all of the features and definitions pertaining to biocontainment laboratories in a chapter dedicated to an overview of design. Therefore, we will concentrate on the elements of building design for "maximum containment" or BSL-4 facilities. 

Microbiological laboratories are divided by the BMBL into four different classifications or levels, with level 1 intended for work with the lowest risk and level 4 designated for work with the highest risk. An ordinary laboratory would approximate a level 2 facility.
