Beckman fragmentation

An intramolecular photocyclization of eucarvone (54), which contains all of the required carbon atoms, was used by Ayer and Browne (96) to give cyclopentanone (55) (Scheme 11). Transposition of the keto group, Beckman fragmentation of the resultant oxime (58), hydrolysis and reduction completed the synthesis. [Pg.21]

The soluble tryptic peptides of 130 mg a chain of Hb-St. Claude were separated on 0.9 x 60 cm columns of Chromobead resin type P (Technlcon Instruments, Dowex 50-X4) at 37°C using the procedure described earlier (16). The method uses a gradient of volatile pyrldlne-acetlc acid buffers of differing molarities and pH as follows first gradient, 666 ml 0.1 M, pH 3.1, and 333 ml 1.0 M, pH 5.0 and second gradient, 166 wl 1.0 M, pH 5.0, and 332 ml 2.0 M, pH 5.0. The amino acid composition of Isolated fragments was determined with a Splnco model 121 automated amino acid analyzer (Beckman Instruments)... [Pg.37]

A thermal cycler (see 8.2.3.2 Polymerase Chain Reaction) is required for the cycle sequencing reaction. MRC Holland offers kits for different sequencing instruments. For more detailed instructions refer to the appropriate instrument user s manual. MLPA fragment analysis requires a fluorescent DNA analysis system. We recommend instruments from Applied Biosystems (e.g. the 16-capillary ABI PRISM 3100 Genetic Analyser) other instruments are available from GC Healthcare and Beckman. [Pg.827]

Figure 7.11 CE-LIF separation and quantitation of fluorescein-labeled lambda PCR product. A 500 bp product was generated with titrated amounts of lambda bacteriophage template and primers, one of which was fluorescein-labeled. The fragment was analyzed by CE-LIF, and only the fiuor from the PCR product was detected. With increasing amounts of DNA template, peak height and area of the product also increased up to a point, whereupon the PCR plateaued. Note the increase in primer and primer-dimer peaks as template availability decreases. (Reproduced with permission from KJ Ulfelder, Applications Information Bulletin A-1774, 1994. PSI particles were produced by digitonin fragmentation of chloroplasts (7). The measurements were made using a 145 000 X g fraction precipitated after 100 000 x g. SDS-PAG electrophoresis of pigment-protein complexes of chloroplasts and PSI peirticles was performed by the method described (8). The radioactivity of the bands was meeisured on "Rackbeta" scintillation counter (LKB). The densitograms of separated chlorophyll-proteins were recorded by a DU-8 spectrophotometer (Beckman) at 675 urn. [Pg.1750]

In a series of different rearrangements, finally under the conditions of fragmentation of a carbon-carbon bond, the Beckman rearrangement of a bicyclic oxime 60 affords cis-chrysanthemonitril 61 (Reaction scheme 38) [68]. [Pg.18]
