Assay antibacterial

Fig. (1). Peptidomics strategies used to study Drosophila immunity. (A) Using antimicrobial assays (antibacterial and antifungal), the bioactive peptides were isolated from the blood of bacteria-challenged Drosophila. MS was used for molecular mass assignment, to identify post-translational modifications, and for primary structure elucidation (B) Identification of peptidic immune effectors through differential display analysis (DD) by MALDI-MS and micro/nano RP-HPLC coupled (online) or not (off-line) to ESI-MS. When the HPLC was performed off-line to the mass spectrometer, fractions were individually analyzed by MALDI-MS. The identification and the structural characterization were performed either by molecular mass assignment and/or sequencing by ESI-MS/MS. Fig. (1). Other specific discovery assays have been used such as differential inhibition of a vancomycin resistant S. aureus strain and its susceptible parent, and an assay based on antagonism of the antibacterial activity by N,A/-diacetyl-L-Lys-D-Ala-D-Ala [24570-39-6] a tripeptide analogue of the dalbaheptides receptor. AppHcation of this latter test to 1936 cultures (90) led to the isolation of 42 dalbaheptides, six of which, including kibdelin (Table 3), parvodicin (Table 3), and actinoidin A2 (68) were novel. A colorimetric assay based on competition between horseradish peroxidase bound teicoplanin and the... [Pg.535]

The results of enzymatic and antibacterial assays demonstrated that compound 165 (as a racemic mixture) exhibits activities similar to those of the parent oxazoline L-159,692 [103]. In order to identify the enantioselectiv-ity of this compound, the corresponding R and S enantiomers were separately synthesized from (R)- or (S)-3-(4-methoxyphenyl)-5-hydroxymethyl-4,5-dihydroisoxazole (166) according to the method of Ukaji et al. [107,108] and tested for LpxC inhibitory and antibacterial activities [103]. Interestingly, only the S enantiomer of 166 exhibited strong enzyme inhibitory and antibacterial activity, while the R enantiomer of L-159,692 was the active component. The enantiomeric synthesis of compound 165 is depicted in Scheme 33. [Pg.212]

Microbiological assay procedures for nalidixic acid have also been used for biological samples. Since nalidixic and hydroxynalidixic acids have the same order of antibacterial activity in-vitro, then cannot be determined separately. [Pg.394]

Compound 10 was evaluated for anti-bacterial activity and acetylchohnesterase (AChE) inhibitory activities. This compound was foimd to be inactive in antibacterial assay and exhibited AChE inhibitory activity with an ICj, value of 67 pM. Acetylcholine serves as a neurotransmitter in the central and peripheral nervous system. Acetylcholinesterase (AChE) stops the function of acetylcholine by its... [Pg.59]

The approach was then widened to include eight vancomycin components, which would dimerise to afford 30 distinct dimers. MS analysis established that 6-(LeuNCH3)C2-(LeuNCH3)C2 underwent the highest amplification. A wide selection of dimers was resynthesized and assayed for antibacterial activity, with excellent correlation being observed between the trends of amplification and activity. [Pg.73]

The third milestone in the history of the use of fermentation products in agriculture was the discovery of the avermectins. They were first detected in an anthelmintic assay using mice infected with nematospiroides dubius (13). This is one of the few assays in which they could have been detected since they lack antibacterial and antifungal activity. [Pg.68]

A number of approaches have been used to discover new antibiotics in past decades. One approach is empirical screening, in which a whole cell assay is used to find antibacterial activities first, followed by the... [Pg.369]
