Antibody gangliosides

A brief discussion of peripheral nerve gangliosides is necessary for understanding the pathogenetic effects of anti-ganglioside antibodies. Gangliosides, the target antigens of... [Pg.270]

Fig. 8.7 CTB-GM1-ganglioside binding ELISA assay. Plates, coated first with GMrganglioside and bovine serum albumin (BSA), respectively, were irrigated with total soluble plant protein from chloroplast transgenic lines (3 and 7) and 300 ng of purified bacterial CTB. The absorbance of the GM1-ganglioside-CTB-antibody complex in each case was measured at 405 nm. Total soluble protein from untransformed plants was used as the negative control. Ellipsometry can follow the interactions between two types of biological macromolecules, the first of those two bound physically to the surface, the other acting from the solution. The binding of conconavalin A to adsorbed mannan 180) and of cholera toxin to adsorbed ganglioside t83) are examples. The adsorption of complement factors to an antibody-coated surface was monitored by ellipsometry and a modification of the same method was used for quantification of migration inhibition of human polymorphonuclear leucocytes 182). Interaction of proteins and cells with affinity ligands covalently coupled to silicon surfaces has been also studied 183). [Pg.54]

Marcus et al. reported that anti GM1 ganglioside antibody can be used as a T cell marker (11, 12, 13). In collaboration with Tada s group of the University of Tokyo we very recently found that antibody to asialoganglio-side GM1 specifically labels mouse natural killer cells and that the antibody selectively kills the cells in the presence of complement (14). Natural killer cells are known by their specific ability to attack tumor cells. We also have evidence that certain gangliosides are capable of eliciting autoimmune diseases experimentally which we called the ganglioside syndrome (JJ5). For example, some brain gangliosides produce autoimmune lesions in peripheral nerve in susceptible rabbits. This may correspond to the human... [Pg.441]

We also investigated the specificities of purified antibodies against G i and Gp, from the serum of a rabbit immunized with bovine brain gangliosides. As shown in Figure 6, the whole antiserum had substantial activity both against the two ganglio-... [Pg.467]
