A novel fiber optic sensor concept using antibody-antigen reactions at a glass-liquid interface was reported by Daehne146. The reaction of antibodies immobilized onto the surface of fused silica fiber optic or planar waveguides with antigens in solution was detected by interaction with the evanescent wave. By detecting in-line fluorescence, the measurement of human IgG is described.

In the most comprehensive structural analysis to date, Lo Conte et al.11 studied 75 protein-protein complexes comprising 24 protease-inhibitor, 19 antibody-antigen, and 32 other complexes (including 9 further enzyme-inhibitor and 11 signal transduction complexes). The authors found that protein-protein interfaces typically have a size of 1600 400 A2 with a few complexes exhibiting very large (2000—4660 A2) or very small (less than 1000 A2) interfaces. Antibody/antigen complex itself may not adsorb and activate factor XII (31), but its formation leads to activation of complement factor Ce which in turn may activate clotting (40, 41). Here too, some products appear poorly soluble and may offer physical interfaces rather than chemical activity.
