Although numerous research articles have been written describing the preparation and use of antibody conjugates with P-gal, the enzyme remains a minor player in ELISA procedures. Less than 1 percent of all commercial ELISA products utilize this enzyme. 

Secondary antibodies incubate sections for 60 min at room temperature with a mixture of secondary antibodies conjugated with different enzymes (e.g., HRP and AP) and raised against the corresponding species IgG or IgG isotypes. Wash sections in a buffer recommended for developing the first enzyme label. The more stable HRP label should be developed before AP. 

The ability to incubate a AAM-NIPAAM copolymer-antibody conjugate with the sample at 37 C should further improve this advantage.

BrdU/DNA flow cytometry offers flexibility and diversity in the study of cell kinetics from cells in culture to human tumors in vivo. The essence of the procedure is to pulse label with BrdU by a short-term incubation in vitro or by a single injection in vivo samples are then taken at time intervals thereafter and stained after fixation in ethanol. The cells are then stained with a monoclonal antibody against BrdU that can be either directly conjugated to a fluoro-chrome (usually fluorescein isothiocyanate [FITC]) or, alternatively, bound to a second antibody conjugated with FITC. The cells are then counterstained with propidium iodide (PI) to measure the DNA content and analyzed on the flow cytometer. The results are displayed as linear-red fluorescence on the x-axis vs linear or log-green fluorescence on they-axis. 

Antibody conjugates with GO can be made using the cross-linking agents glu-taraldehyde (Chapter 10, Section 1.2) or SMCC (Chapter 10, Section 1.1). The heterobifunctional reagent SMCC provides the best control over the conjugation process and usually results in high-activity preparations.

Horseradish peroxidase is one of several enzymes that are commonly used for the detection of antigens in the ELISA. Like HRP-conjugated antibodies, secondary antibodies conjugated with these other enzymes are commercially available. A list of ELISA detection enzymes, the substrates that they use, and the wavelengths of detection for the soluble colored products that they produce is presented in Table 17-1.

The fluorochromes, Calcofluor White and Congo red bind to chitin and cellulose, while acridine orange binds to nucleic acids of the fungi. These dyes may be helpful when there is a lack of fungal elements (blastospores, hyphae or pseudohyphae) in the specimen, as even few elements are visible under the fluorescence microscope. By using specific antibodies conjugated with the fluorochromes, it should be possible to identify specific fungi.
