The thalidomide analogs CPS49 (30) and CPS11 (31) have been reported to inhibit PI3/AKT signaling in multiple myeloma cells via an anti-angiogenic effect. These compounds are devoid of the teratogenic properties seen with thalidomide and are currently in preclinical development [66]. Compound 30, and to a lesser extent 31, induced a dose-dependent inhibition of proliferation in several multiple myeloma cell lines and reduced phospho-AKT levels [66]. These compounds also inhibited DNA synthesis in cell lines resistant to conventionally used anti-multiple myeloma drugs (e.g. dexamethasone, anthracyclines and melphalan) in a dose-dependent manner. [Pg.373]

This chapter describes the metabohsm of sphingosine and SIP, evidence to support their pro- and anti-apoptotic effects and their proposed sites of action on signaling processes that contribute to apoptosis and proliferation. [Pg.246]

Almost 20 years ago the ammonium tellnrolate AS 101 was demonstrated to possess immnnomodnlating properties and to mediate anti-tumour effects in rats. The same com-ponnd stimnlates hnman lymphoid cells to proliferate and produce lymphokines, tumour necrosis factor (TNF) and other cytokines in vitro ... [Pg.333]

These novel tetrameric bromotyrosine metabolites display a range of biological activities, including effects on calcium channels (1900), lipoxygenase inhibition (1772), tumor angiogenesis inhibition (1901), and endothelial cell anti-proliferation (1902). Syntheses of several bastadins have been accomplished (1903,1904). [Pg.314]

Aspirin, now commonly used for prevention of CHD and colon cancer, combined with butyrate exerted stronger anti-proliferate and pro-apoptotic effects than either alone. [Pg.628]
