In another extensive series of studies, Clarke and coworkers developed sensitive microchemical tests for the determination of alkaloids [32-34], anesthetics [35], antihistamines [36], antimalarials [37], and analgesics [38]. One of the useful techniques introduced by Clarke was that of the hanging-microdrop [32], which permitted identification tests to be made on submicrogram quantities of analyte. Results obtained on the cinchona alkaloids are shown in Table 5 to illustrate the methodology. 

With the development of chemistry in the early 1800s came the understanding that natural products owe their medicinal properties to certain substances they contain. In 1806, for example, morphine was isolated from opium, and in 1820 quinine, a drug useful in fighting malaria, was isolated from the bark of the cinchona tree. Soon, compounds produced in the laboratory were also found to have medicinal properties. In the 1840s, for example, anesthetic activity was found in the synthetic chemicals chloroform, nitrous oxide, and ethyl ether, making painless surgery and dentistry possible.

Anesthetic effects have been observed with some of the cinchona alkaloids. Morgenroth and Ginsberg (34) applied some of these products to the cornea of rabbits for 2-8 min. A normal anesthesia was considered as a 30-90-min. suppression. Normal anesthesia was obtained by a 3% solution of quinine hydrochloride and by a 1-1.25% solution of hydroquinine and ethylhydrocupreine hydrochloride. 

The principal action of the quaternary cinchona alkaloids has been shown to be peripheral (50, 87, 88). The central nervous system is not markedly effected by paralyzing doses, but a transient fall in blood pressure is produced (50, 87, 88, 104). A -Methylquininium chloride has been used on a limited scale in clinical medicine to relieve muscle spasm in certain spastic states (5a, 9, 31). As in the case of curare and the Erythrina alkaloids, synergistic effects are noted between iV-methyl- and iV-ethylquininium chlorides and certain anesthetics and hypnotics (45).

Several alkaloids are used in medicine. Quinine, obtained from the bark of the cinchona tree, has been used in the treatment of malaria since the 1600s. Atropine from nightshade (belladonna) is used in low concentrations to accelerate slow heart rates and as an anesthetic for eye examinations. 

Cinchona has astringent and bitter tonic properties also reportedly has analgesic and local anesthetic properties, among others (Goodman AND Gilman jiangsu naniing).
