Several studies were performed on the optimization of expression levels of ELP proteins in E. coli. In a recent example, the expression protocol was optimized for an ELP fusion with green fluorescent protein (GFP). This fusion protein was expressed and purified in a yield of 1.6 g/L of bacterial culture, which finally yielded 400 mg GFP/L bacterial culture. This extremely high yield was found after uninduced expression in nutrient-rich medium supplemented with phosphate, glycerol and certain amino acids, such as proline and alanine [234]. The influence of fusion order was also examined and it was found that positioning the ELP at the C-terminus of target protein resulted in significantly higher expression levels [35]. [Pg.80]

The answer is B. While all of the listed conditions are consistent with lethargy and developmental defects, the lactic acidosis rules out pyruvate kinase deficiency. Thiamine and niacin deficiencies are unlikely due to the lack of effect of vitamin supplementation. Excess pyruvate is the source of the elevated alanine in the serum. The clinical findings are thus consistent with pyruvate carboxylase deficiency, which is associated with severe hypoglycemia due to fasting due to impaired gluconeogenesis. [Pg.101]

Pantothenic acid is produced commerdally by synthesis involving the condensation of d-pantolactone with salt of -alanine. Some of the dietary supplement forms include caldum pantothenate, dexpanthenol, and panthenol. [Pg.1204]

L-Aspartic acid (12) is an industrially important, large volume, chiral compound. The primary use of aspartic acid in the fine chemical arena is in the production of aspartame, a high potency sweetener (Chapter 31). Other uses of L-aspartate include dietary supplements, pharmaceuticals, production of alanine (by decarboxylation), antibacterial agents, and lubricating compounds. There have been a number of reviews on L-aspartate production.53 56... [Pg.24]

Certain isolated amino acids L-glutamine, L-alanine, and glycine when taken together as a supplement have shown beneficial activity in reducing the symptoms of BPH. [Pg.83]

Fig. 7-19 and Fig. 7-20 show a number of effects on the activity of aminoacylase, which cannot be described by a standard kinetic model alone. To derive a kinetic model for the aminoacylase and for describing all influences, a reaction scheme based on the rapid equilibrium random uni-bi model was used (compare with Eq. (40)). This model was supplemented, according to the above measured influences of substrate inhibition by acetate and inhibition by D-alanine. Consequently, the reaction scheme Eq. (40) had to be extended to yield equation (46) and the corresponding rate equation (47), which is modified by the additional equilibria ... [Pg.227]

Various sugars make the listings, denoted by the suffix -ose. Various amino acids also make the listings, denoted by the suffix -ine, notable examples being L-alanine and L-phenylalanine, commonly associated with nutritional supplements. Arginine, an essential amino add (not manufactured in the body), is said to be an inhibitor. The suffix pyridoxal is fisted as an inhibitor, whereby it may be noted that pyridoxine (or pyridoxin) is called vitamin B6. [Pg.105]
