The third part. Developmental Psychopathology, delves into detailed disease-specific overviews. Each of the chapters covers issues pertaining to nosology and classification, to genetic determinants, brain systems implicated, environmental influences, and nature-nurture interactions. Neurotransmission and neuromodulation, and hormonal and other developmental influences are addressed and, whenever available, relevant animal models are incorporated into the discussion. The interplay of normative and derailed development is a core concept for these chapters. Of the part s 12 chapters, 9 are devoted to traditionally defined disease categories, and 3 cover the overarching areas of early-life stress, aggression, and affiliative behaviors. [Pg.813]

The two other genes clearly linked to AD are presenilins (PS) 1 and 2 (37,38). Missense mutations in these homologous 8-TMD polypeptides cause the most aggressive form of AD known, with onset commonly occurring in the 40s and 50s and very rarely as early as the late teens. Many studies in cell culture, animal models, and patients demonstrate that presenilin missense mutations elevate the y-secretase-mediated production of the strongly self-aggregating A/142 peptide, perhaps at the expense of the A/i40 peptide (14). The antidepressant activity of hypericum has been extensively investigated over the last two decades in animal models (forced-swimming and tail-suspension tests) as well as in humans. Clinical trials have demonstrated an improvement in symptoms of anxiety, dysphoric mood, hypersomnia, anorexia, depression, insomnia, psychomotor retardation, and other subjective indicators.Potential for the treatment of premenstrual syndrome (PMS) also exists. Earlier studies also showed that hypericum enhanced mice exploratory activity in a foreign environment, extended narcotic sleeping time (dose dependent), is a reserpine antagonist, and decreased aggression in socially isolated male mice. Hypericin has been found to inhibit in vitro almost irreversibly both type A and B monoamine oxidase (MAO) in rat brain mitochondria.
