Methyl-a-D-glucoside

When dextrose is heated with methanol containing a small amount of anhydrous hydrogen chloride, a-methyl-D-glucoside is obtained in good yield and can be isolated by crystallization. Similar reactions occur with higher alcohols, but the reaction products are more difficult to isolate by crystallization. Dextrose reacts with acid anhydrides in the presence of basic catalysts, yielding esters. Complete reaction gives the pentaacylated derivative. 

A stereocontrolled route to the native form of thromboxane B2 was carried out starting with a-methyl-D-glucoside (Ref. 3).

The stereoselective requirements for the transport of sugars by these transporters are quite restrictive. The D isomers of mannose, fructose, and galactose appear to interact with both systems however, only D-galactose, a methyl-D-glucoside, and 6-deoxy-D-glucose appear to be transported... 

Treatment of a methyl D-glucoside with methyl sulfate and sodium hydroxide brings about methylation of the four remaining —OH groups, and yields a methyl tetra-O-methyl-D-glucoside ... 

The differential titration curves are shown in Fig. 1. As reported (29), the first peak on each curve reflects the neutralization of the OH group at C2. A comparison of the peaks shows that the degree of dissociation of the OH group at C2 is lower in a-methyl-D-altroside than in a-methyl-D-glucoside.
