A new antiviral agent, developed for treatment of CMV retinitis, can be administered by intravitreal injection. Currently, there is only one antisense drug on the market— Vitravene (active ingredient fomivirsen) for the treatment of cytomegalovirus (CMV)-induced retinitis (inflammation of the retina) in AIDS patients. Fomivirsen has 21 nucleotides complementary to a CMV mRNA sequence, which is necessary for the production of infectious virus. Two examples of experimental antisense drugs are provided in Exhibit 3.15, while Table 3.1 lists other antisense drugs in clinical phase. [Pg.81]

Recommended dosage and monitoring requirements For the treatment of CMV retinitis in AIDS patients, the recommended dose of Vitravene is 330 pg (0.05 ml) by intravitreal injection for induction, one dose should be injected every other week for two doses for maintenance, after induction, one dose should be given once every 4 weeks after induction. [Pg.333]

Drug interactions Vitravene is not recommended for use in patients who have recently (2-4 weeks) been treated with either intravenous or intravitreal cidofovir because of the risk of exaggerated ocular inflammation. Results from in vitro tests demonstrated no inhibition of anti-CMV activity of fomivirsen by zidovudine or zalcitabine. [Pg.333]
