Floyd et al. (F8a) were the first to present in vitro evidence for DNA alterations by a free radical-generating tumor promotor when they showed that 8-hydroxyguanine was a product of oxidatively damaged DNA. 

Chlormethine can act as a tumor promotor. In most cases squamous cell carcinomas and basal cell carcinomas have been reported. Two cases of small malignant melanoma, 3 mm in diameter, have been reported in patients with mycosis fungoides stage la, with latency intervals of 18 and 10 months after withdrawal of local application, which had been conducted for 18 months and almost 3 years respectively (20).
