Tryptophan adverse effects

The possible antimanic effect of this 5-HT precursor was postulated based on the permissive hypothesis concept of diminished 5-HT activity. When oral doses of L-tryptophan (1 to 4 g) are administered, there is evidence of increased 5-HT synthesis. Three of four double-blind studies yielded positive results, holding the promise of an effective antimanic treatment (275). An advantage of this drug is its relative lack of other adverse effects. 

The syndrome appears to be only part of a spectrum of adverse effects associated with tryptophan (15). There has been much discussion, but finally it appears that the links are causal, as consistent findings were found in multiple independently conducted studies and the incidence of eosinophilia-myalgia syndrome in the USA fell abruptly once tryptophan-containing products were recalled (16).

Pellagra-associated encephalopathy has been suspected as an adverse effect of isoniazid administration in several patients with tuberculosis. Deficiency of niacin (nicotinic acid) is characterized by dermatitis, diarrhea, and dementia. Other symptoms can occur, such as seizures, hallucinations, spasticity, and glossitis. Pellagra induced by isoniazid is promoted by malnutrition or a vegetarian diet with low intake of the nicotinamide precursors tryptophan and nicotinic acid. Specific supplementation is essential (10). 

In humans, few adverse effects have been reported with L-tryptophan, even when taken in large doses (up to 9 g/day) for extended periods of time. Even though tryptophan was used clinically for over 20 years, the literature had few reports of adverse effects when pure tryptophan supplements were given to humans.5 Thomson et al.6 reported that tryptophan alone (3 g/day) produced no more side effects than did placebos. In studies using higher doses of tryptophan (9.6 g/day) in affective disorder patients7 and 20 g/day in schizophrenic patients,8 no side effects were reported. In other studies where side effects were found, they were mild (most common being nausea and lightheadedness).5... 

Adverse effects of mercury to fishes, in addition to those listed on reproduction and growth, have been documented at water concentrations of 0.88-5.0 xg/L enzyme disruption in brook trout (Salvelinus fontinalis) embryos immersed for 17 days in solutions containing 0.88 ig/L, as methylmercury decreased rate of intestinal transport of glucose, fructose, glycine, and tryptophan in the murrel (Channa punctatus) at 3.0 (xg Hg +/L for 30 days altered blood chemistry in striped bass (Morone saxatilis) at 5.0 xg Hg +/L in 60 days and decreased respiration in striped bass 30 days post-exposure after immersion for 30-120 days in 5.0 xg Hg +/L. In large-mouth bass, elevated liver metallothioneins are indicative of elevated muscle mercury concentrations, suggesting that mercury-induced metallothioneins may be useful biomarkers of mercury exposure. 

This is a larger variety of millet widely grown in Africa and Asia. Its protein contains less of the S-containing amino acids and it has a high leucine content which may have an adverse effect when intakes of tryptophan and nicotinic acid are low.
