Thiogalactopyranosides

X stock solution of isopropyl / -D-l-thiogalactopyranoside (IPTG) (0.4 m) 1 g IPTG dissolved in 10 ml bidest water sterilize by filtering through a 0.22 pm filter 1000 X stock solution of ampicillin (50 mg ml ) 1 g ampicillin dissolved in 20 ml sterilized bidest water... 

The culture was charged with 1 mM isopropyl-/3-D-thiogalactopyranoside to induce gene expression and cultivated another 3 h. 

Biological. Dibenzofuran was oxidized by salicylate-induced cells of Pseudomonas sp. strain 9816/11 and isopropyl-P-D-thiogalactopyranoside-induced cells of Escherichia co//JM109 (DE3) (pDTG141) to (li ,25)-ds-l,2-dihydroxy-l,2-dihydrodibenzofuran (60-70% yield) and 3SARj-ds-3,4-dihydroxy-3,4-dihydrodibenzofuran (30-40% yield) (Resnick and Gibson, 1996). 

Effect of the addition of isopropyl- 3-D-thiogalactopyranoside (IPTG) (USB) at set cell densities (OD660between0.01 and 0.8) corresponding to cultures at 103-107 CFU/mL. 

Concentrated PBS (10X), 0.5 Af ethylendiamine tetraacetic acid (EDTA), 1 Af Tris-HCl pH 8.0, 5 Af NaCl, 3 AfNaAc pH 5.2, kanamycin and 1 M isopropyl-(3-D-thiogalactopyranoside (IPTG) from Gibco/BRL (Bethesda, MD).

GST-C3 fusion protein expression is induced by adding isopropyl- 3-D-thiogalactopyranoside (IPTG Calbiochem) to 0.2 mM (1 ml of 0.1 M stock made in water and stored at -20 "C), and the culture is incubated with shaking for a further 3h. 

Isopropyl P-3-thiogalactopyranoside (IPTG) (Life Technologies) 100 mg/mL in water. Sterilize by filtration through a 0.22-p disposable filter. Store in aliquots at -20°C.
