Standard Staining Protocol ABC Method

Immunohistochemistry is a powerful and very useful tool in diagnostic pathology consequently, the staining protocol must be optimized to ensure that the results are reproducible, reliable and accurate. A standard immunohistochemistry method includes the following steps  

Cutting optimal paraffin sections, mounting on adhesive slides and drying. 

Elimination of non-specific background by incubation with non-immune serum. 

Reaction with a biotinylated secondary (link) antibody. 

Developing the reaction product with chromogen, counterstaining, dehydration, covering, and interpretation.
