Oligomeric deoxyguanosines 69 all assemble into columns in water.93 Using SANS, the average length of the columns (DP) of freshly prepared samples was determined to be around 6 nm at a concentration of 1% in D2O.88 Increase of the temperature resulted in the loss of the cylindrical aggregates. When the sodium salt was used, subsequent cooling did not result in the reformation of the cylindrical polymers. In contrast, the potassium salt did allow for reversible self-assembly. The addition of extra potassium salt even increased the DP. It binds to the inner carbonyls of the G-tetramer and stabilizes the tetramer 94... 

The same group also performed experiments with a continuous chromatographic reactor of the SMBR type [133,174]. The system used consisted of 12 columns with inner diameters of 5.4 cm and a length of 75 cm. Approximately 12 cycles were necessary for stabilization of the system. Complete inversion of sucrose even at feed concentrations of up to 55% w/v was achieved, while the product purity was over 90%. After prolonged usage, the system efficiency decreases due to the loss of the calcium ions from the matrix and a consequent decreased selectivity of the resin. However, this problem could be overcome by regeneration of the resin with calcium nitrate. 

To test for evaporation, the weight loss should be determined on suitable aliquots of a test solution stored in the automated workstation under typical environmental conditions for the length of the desired stability before reassaying. When compared to the initial weight of the test solution, the weight change should not be greater than 2.0% and the response of the solution should be between 98.0 and 102.0% of the initial value. 

Each of these elements is a critical component of the stability program, but the proper operation of environmental chambers is one element that is most often assumed. It is often taken for granted that in the absence of external factors such as power loss or mechanical failure, a chamber, will maintain its predefined storage condition over the length of the stability study, but this is rarely the case. How do we ensure that the chamber will perform and maintain its set-point conditions as expected over the course of the stability study A properly executed qualification program of the chamber prior to its routine use in a stability program will verify that the chamber can perform as expected. 

In general, a drug product should be evaluated under storage conditions (with appropriate tolerances) that test its thermal stability and, if applicable, its sensitivity to moisture or potential for solvent loss. The storage conditions and the lengths of studies chosen should be sufficient to cover storage, shipment, and subsequent use.
