Specialized scale-up culture systems

An extension of the flask culture principle for ving lai e unit surface areas arc the Nunc A/S Cell Factories (16). These are polystyrene plates of 632 cm which are available in single, double (1264 cm ), 10 Tray (6320 cm, and 40 tray (25280 cm ) units. Each tray level requires 200 ml medium and can be filled, harvested, etc. in one operation per factory unit. For large scale commercial processes the system can be semi-auromated with the Manipulator and Robot System. 

Growth of hybridoma cells In Nunc Cell Factories 

Prepare Cell Factory in a Class II cabinet by removing packaging and fixing the provided air filter and tube connector to the two open adaptor caps. 

Prepare cell suspension in aspirator bottle (2 litres for a 10 tray unit with 5-10 X 10 cells homogeneously mixed) and connect to the tube connector in the Cell Factory. 

Turn Cell Factory on its side with the supply tube at the bottom, and with the aspirator above the Cell Factory open the clamp and allow cell suspension to flow in gently under gravity (alternatively a minimal amount of air pressure may be applied using a pump).
