Size-exclusion chromatography hyphenated techniques

The complementary use of LC-ESI-ToF-MS and hyphenated ICP-MS with dynamic reaction cell (DRC) for the characterization of native and recombinant copper proteins (of molar mass range 10-20kDa) has been investigated by Hann et al.67 Size exclusion chromatography (SEC) and ion chromatography (IC) were implemented as separation techniques for hyphenated ICP-MS of intact Cu containing proteins. 

The follow-up section will deal with separation methods based upon (a) molecular size and related to it hydrodynamic volume (size-exclusion chromatography and ultrafiltration), (b) molecular size and related to it molecular diffusivity (field-flow fractionation), and (c) charge/size ratio and related to it molecular polarity (electrophoresis and mass spectrometry). Also reviewed will be hyphenated techniques or those that combine separation by chromatography or electrophoresis with spectral detection. 

Anion exchange chromatography coupled with ICP MS was used in the simultaneous speciation analysis of As, Se, Sb and Te compounds in extracts of fish [230]. Size exclusion chromatography (SEC) coupled with specific detectors is frequently used to analyse species of trace elements in protein-rich materials, such as extracts of meat and plant tissues. For instance, SEC hyphenated with ICP MS was used for the speciation analysis of Cu and Zn in samples of leguminous plants [191]. The same technique was applied to the speciation analysis of Cu, Cd, Zn, Se, As and Ca in fish [220] and Fe, Zn, Cu, Ag, Cd, Sn and Pb in mussels [189]. SEC HPLC coupled with GF AAS mmed out to be very useful for determining levels of Fe species in baby food [312]. With gel permeation chromatography (GPC) GF AAS, the speciation forms of Cd were determined in two kinds of vegetables contaminated with this element [216]. 

This book covers some of the significant advances in hyphenated chromatographic separation methods for polymer characterization. Chromatographic separation techniques in this volume include size-exclusion chromatography, liquid chromatography, and field flow fractionation methods that are used in conjunction with information-rich detectors such as molecular size-sensitive or compositional-sensitive detectors or coupled in cross-fractionation modes.
