The genotoxic effects of nalidixic acid (400 mg bd for 10 days) and metronidazole (250 mg tds for 10 days) have been investigated in a prospective randomized study in 20 patients with Trichomonas vaginalis infections (28). Evaluation was by the sister-chromatid exchange test, in which an increased number of exchanges in lymphocytes reflects mutagenic action. Metronidazole had no effect but there was a significant increase with nalidixic acid. 

Propionic acid has been tested using the Escherichia coli DNA repair assay, the SOS chromotest, the Sal-monella/microsome mutagenicity test, and the sister chromatid exchange test in vitro. All tests except the DNA repair assay with E. coli yielded negative results. These data support other evidence that propionic acid is not mutagenic and that genotoxic events are unlikely to be the cause of forestomach lesions in rats fed propionic acid in the diet.

Although teratogenicity was seen in chick embryos (Hall 1976), results in rats, mice, and cats were conflicting (Manzo and Sabbioni 1988), with both Ames and Sister chromatid exchange tests proving negative (Claussen et al. 1981).
