Signaling aptamers

Hamaguchi et al. (2001) have constructed a thrombin detection system using a designed thrombin aptamer and fluorescence resonance energy transmission (FRET). The oligonucleotides complementary to the nucleotides at the 3 end [Pg.131]

These signaling aptamers have shown that multiple aptamer ligands can be detected without bound-free separation that is, through homogeneous detection. It is very difficult to realize this type of detection using antibodies, aside from a few exceptions (Ueda et al., 2003). We discuss the problem of homogeneous detection in the next section and introduce our solution the aptameric enzyme subunit. [Pg.132]

Various optimizations were carried out to improve the cell-specific signal. Aptamer-probe concentrations ranging from 1 nM to 0.1 pM were attempted in the presence of a minute concentration of splint (400pM Fig. 2). The same probe dilution series was also performed in the presence of even smaller splint concentrations of 40pM and 4pM (Fig. 3). Overall, it is difficult to... [Pg.390]

Figure 36 The structure-switching signaling aptamer. A DNA duplex composed of three strands of DNA places a fluorophore (F) close to a quencher group (Q). Upon addition of thrombin, the QDNA piece is released, and the fluorescence increases (See ref 67). Figure 36 The structure-switching signaling aptamer.
