Serological cross reactivity

The 8-O-acetyl-associated epitope of legionaminic acid was also found in P. fluorescens ATCC 49271, which has the same OPS structure 17 as L. pneumophila serogroup l32,56 (Fig. 5). This epitope, possibly along with other, yet uncharacterized cross-reactive epitopes, is likely responsible for the serological cross-reactivity exhibited by P. fluorescens and distantly related L. pneumophila.

For demonstrating serological cross-reactivity it is desirable to immunize a recipient animal which is not closely related to the two species whose immunoglobulins are to be compared. For example, Orlans and Feinstein immunized chickens with mammalian IgA proteins in order to demonstrate cross-reactivity (20). It is obvious that one should not use as recipient a species closely related to the donor since it may not recognize as foreign many of the determinants of the injected molecule. 

All current data support this duplication hypothesis. These include detailed serological analyses skin grafting experiments , and an extensive series of experiments to directly test the postulate of serological cross-reactivity between the products of the H-2K and H-2D genes . Most importantly, further biochemical analyses of the H-2 antigens have entirely supported the hypothesis. 

Another indicator that the SEs share similar structures is evidenced by cross-reactivity and neutralization with antibodies. Several years ago, when there were less SEs known, these molecules were considered serologically distinct entities as determined by antisera and relatively insensitive immunodiffusion assays. However, subsequent studies employing a more sensitive technique (ELISA) with polyclonal and monoclonal antibodies clearly reveal that common epitopes do indeed exist between these toxins.
