Conformational epitopes will be lost in many instances but it is unlikely that a relevant antigen will not be detected for this reason in the current immunoblot analyses, since most immunogenic proteins induce polyclonal responses that likely recognize conformational and sequential epitopes.

From the 11 epitopes tested, 3 appeared to be immunodominant. Hazelnut is known to cross-react with tree pollen and only little data exist on pollen-unrelated allergens causing hazelnut allergy. As one example, the cDNA of Cor a 9, an 11S globulin seed storage protein has been cloned, but no experimental data on sequential IgE-binding epitopes have been obtained (Beyer et al., 2002). 

As many B cell epitopes are described to be conformational (Aalberse, 2000), peptides identified by sequential approaches may render limited information and might exhibit reduced antibody-binding affinity. In a very stringent definition, conformational epitopes are composed of amino acids from different loci in the allergen sequence brought into proximity by the tertiary structure of the protein. Systematic characterization of conformational epitopes is much more difficult compared to linear epitopes, because it requires knowledge of the 3D structure, which is often limited. 

Within the last years many food allergens have been mapped, and the majority of sequential IgE-binding epitopes were found. These data have been feeding evergrowing databases (Korber et al., 2006) and can be used in bioinformatic approaches...
