Triethylamine and sec-butyl chloroformate added dropwise at -15° to a stirred ethereal soln. of N-benzyloxycarbonyl-L-proline, the resulting triethylamine hydrochloride removed by filtration, ethereal diazomethane added dropwise, stirred 4 hrs. at 0°, and allowed to stand overnight in a refrigerator a-diazoketone (Y 91%) dissolved in methanol, and refluxed 12 hrs. with AggO on a water bath -> methyl N-benzyloxycarbonyl- -L-homoprolinate (Y 95.1%). It is significant that under these conditions, with unactivated alcohols, chloro-formates are isolated readily without a trace of the corresponding chloride (—)-menthol chloroformate and sec-butyl chloroformate were prepared in yields of 98% and 78%, respectively.

In each solvent series,the partition coefficient of the sample can be finely adjusted by modifying the volume ratio of the components. The first series covers a broad range in both hydrophobicity and polarity continuously from M-hexane/methanol/water to n-butanol/water. The second series of chloroform/methanol/water provides moderate hydrophobicity and the third series of ferf-butyl methyl ether/w-butanol/acetonitorile/water is suitable for hydrophilic compounds. Most of these two-phase solvent systems provide near 1 1 volume ratios of the upper/lower phases, together with the reasonable range of settling times in 30 sec or less, so that they can be efficiently applied to HSCCC and other centrifugal CCC schemes.
