Sciatic nerve transection results in an increased mRNA concentration and elevated NGF synthesis distal to the lesion (Abrahamson et al., 1987 Heumann et al., 1987a,b). Novel material options are being explored as matrix fdlers in NGCs. Recently, a sericin/silicone-based NGC was fabricated and used as a long-term implant in vivo. Sericin conduit wrapped with a silicone conduit was used for the bridging repair of a 5-mm gap in a rat sciatic nerve transection model. These double conduits achieved functional recovery comparable to autologous nerve graft and, importantly, positive results were mainly attributed to sericin (Xie et al., 2015).

Initial experiments in animals with complete spinal cord transection demonstrated that local application of macrophages that have been co-incubated with sciatic nerve promoted motor recovery (Lazarov-Spiegleretal., 1996 Rapalino et al., 1998). Subsequently, the experiments were repeated in a model of severe spinal cord contusion. In those experiments blood-borne monocytes were activated by co-incubation with autogeneous skin (Bomstein et al., 2003). The results revealed that this was equally effective for recovery from spinal cord injury. In these and subsequent experiments, the macrophages were characterized phenotypically, and parameters such as site of injection, dosing and therapeutic window were studied. Those macrophages were found to have a dendritic-like phenotype (Bomstein et al., 2003) and expressed features that are reminiscent of alternative activation production...

The striking plasticity in Na+ channel expression that can occur in normal neurons raises the question of whether there are alterations in W channel expression in injured neurons. Dorsal root ganglion (DRG) neurons have been especially well studied in this respect. As illustrated in Fig. 5 (middle and bottom rows), following transection of the peripheral axons of DRG neurons (by ligation of the sciatic nerve), expression of mRNA for the SNS (Dib-Hajj et al 1996) and NaN (Dib-Hajj et al 1998a) Na channels is down-regulated. The reductions in SNS and NaN transcript expression are paralleled by reduced levels of SNS and NaN Na channel protein (Sleeper et al 2000).
