I been established to serve as a registry of convicted offenders. When a DNA sample is obtained from a crime scene, the sample is subjected to cleavage with restriction endonucleases to cut out fragments containing the STR loci, the fragments are amplified using the polymerase chain reaction, and the sequences of the fragments are determined. 

The most recent advances on enzymatic inhibition are related to endonucleases and polymerases. A tris-malonic acid fullerene derivative can interfere with DNA restrictive enzymatic reactions, demonstrating a dose-dependent inhibition of these enzymes, with an IC50 in the micromolar range. The addition of ROS scavenger does not revert the enzymatic activity, indicating that the fullerene action should be exerted in a direct way (Yang et al., 2007d). 

Restriction endonuclease EcoRI is able to cut a chain in dsDNA which has a chiral phophorothioate group at the specific cleavage site.838 Tire reaction occurs with inversion of configuration at phosphorus, suggesting direct in-line attack by a hydroxyl ion generated from H20.

Purify the reaction products by using a PCR purification kit or by ethanol precipitation (see Section 3.3.2 steps 5-10), digest with appropriate restriction endonucleases, and ligate into the cloning vector.
