The Kobayashi group devised a two-step synthesis of 7-aryl-6,7-dihydrothieno[2,3- ]pyrazines. Aryl(3-chloropyrazin-2-yl)methanones were treated with methylene triphenylphosphorane under Wittig reaction conditions to furnish alkene 155, which in turn is stirred with aqueous sodium hydrosulfide, affording 6,7-dihydro-7-arylthieno[2,3-f)]pyrazine 156 (Scheme 77) (13HTC1507). Subsequent dehydrogenation with a catalytic amount of 10% palladium on carbon provided the desired 7-aryl-6,7-dihydro thieno [2,3-b] pyrazine 157. [Pg.438]

Microwave and fluorous technologies have been combined in the solution phase parallel synthesis of 3-aminoimidazo[l,2-a]pyridines and -pyrazines [63]. The ring cleavage of 3-aryl-2-substituted-2//-azirines by molybdenum hexacarbonyl has been described earlier in regard to the synthesis of pyrroles, pyrazoles and isoxazoles. A study of the a-arylation of diazine mono iV-oxides, under Heck-like conditions, also gave emphasis to pyrazines but a number of examples using pyrimidines and pyridazines were also described (Scheme 1). A wide range of aryl chlorides, bromides and iodides was used and the products were easily deoxygenated by catalytic reduction. A different approach to quinoxalines and heterocycle-fused pyrazines has been described by the Lindsley group, based on the cyclocondensation of 1,2-diketones and aryl/heteroaryl 1,2-diamines (Scheme 6.260) [450]. Optimized reaction conditions involved heating an equimolar mixture of the diketone and diamine components for 5 min at 160 °C in a 9 1 methanol/acetic acid solvent mixture, which furnished the substituted quinoxalines in excellent yields. This approach could also be applied equally successfully to the synthesis of heteroaryl pyrazines, such as pyr-ido[2,3-b]pyrazines and thieno[3,4-b]pyrazines. The same group has employed 1,2-diketone building blocks for the preparation of other heterocyclic structures (see Schemes 6.198, 6.268, and 6.269). [Pg.270]

The vinyl triflate of Komfeld s ketone has been subjected to Heck reactions with methyl acrylate, methyl methacrylate, and methyl 3-(Af-rerf-butoxycarbonyl-lV-methyl)amino-2-methylenepropionate leading to a formal synthesis of lysergic acid [259]. A similar Heck reaction between l-(phenylsulfonyl)indol-5-yl triflate and dehydroalanine methyl ester was described by this research group [260]. Chloropyrazines undergo Heck couplings with both indole and 1-tosylindole, and these reactions are discussed in the pyrazine Chapter [261], Rajeswaran and Srinivasan described an interesting arylation of bromomethyl indole 229 with arenes [262].
