Phosphofructokinase (PFK) purified from A. suum muscle is inhibited by ATP and stimulated by AMP, K", NH2, phosphate, and hexose bisphosphate (19). It has been estimated that the A. suum PFK functions at 1-2% of its maximal velocity in vivo. Both phosphorylated and dephosphorylated forms of the A. suum PFK exhibit similar specific activities, but the dephosphorylated enzyme has a 2-2.5-fold lower activity in a physiological assay system. Phosphorylation of the dephosphorylated PFK with the mammalian cAMP-dependent protein kinase results in the incorporation of about 1 mol of phosphate/mol of subunit and completely restores activity in the physiological assay system (20). Therefore, it appears that PFK activity in A. suum muscle can be regulated, not only by metabolites, but also by covalent modification catalyzed by a cAMP-dependent protein kinase. This contrasts with mammalian muscle, where the effect of PFK phosphorylation is poorly defined and ranges from inhibition to mild stimulation (20). [Pg.52]

Abstract— This paper reported an improvement to the previously proposed maximum-minimnm variation (MMV) test for protein estimation. The proposed method ntilizes single protein test as compared to three protein tests for MMV. In addition, a new computerized algorithm namely maximum-minimum area variation (MMAV) has been proposed in order to estimate the protein concentration in the latex glove. The new proposed technique, give significantly better results in terms of consistency and accuracy than existing methods. Biuret method This method is simple and reasonably specific as it depends on the reaction of cop-per(II) with four N atoms in the peptide bonds of proteins. Compounds containing peptide bonds give a characteristic purple colour when treated in alkaline solution with copper sulfate. This is termed the biuret reaction because it is also given by the substance biuret NH2CONHCONH2. For a wide variety of proteins, 1.0 mg of protein in 2 cm of solution results in an absorbance at 540 nm of 0.1. Many haemoproteins, for example, give spurious results due to their intrinsic absorption at 540 nm, but modifications which overcome this difficulty are known (either removal of the haem before protein estimation or destruction of the haem by hydrogen peroxide treatment). The protein content of cell fractions such as nuclei and microsomes can be estimated by this method after solubilization by detergents such as deoxycholate or sodium dodecyl sulfate. [Pg.137]
