The excitatoiy amino acids (EAA), glutamate and aspartate, are the principal excitatory neurotransmitters in the brain. They are released by neurons in several distinct anatomical pathways, such as corticofugal projections, but their distribution is practically ubiquitous in the central nervous system. There are both metabotropic and ionotropic EAA receptors. The metabotropic receptors bind glutamate and are labeled mGluRl to mGluRB. They are coupled via G-proteins to phosphoinositide hydrolysis, phospholipase D, and cAMP production. Evaporate in vacuum to get 3-Br-isoxazole-5-COOH (II) (re-crystallize-benzene-toluene). Dissolve 30.7 g (11) and 27 g KOH in 500 ml methanol and stir two hours at 140°. Cool, add 1.5 L water and extract with 3X1.5 L ether. Can decolorize aqueous solution by boiling briefly with carbon, then filtering and acidifying with concentrated HCl. Filter and recrystallize-benzene to get 3-methoxy-isoxazole-5-COOH, which is the analog of (IV) in method 1 and is identical to (11) in method 3 and can be converted to muscimole by either method. [Pg.100]
