The main advantage of the IPG-Dalt protocol is a much higher reproducibility in gel preparation and running conditions, and mostly higher resolution of the sample.

Preparation of sample and buffers DNA is prepared from the acquired sample using conventional DNA extraction techniques and is mixed with a loading buffer containing bromophenol blue, ready to be loaded into the wells on the agarose gel. Typical buffers used for agarose gel electrophoresis are trisacetate EDTA (TAE) or trisborate EDTA (TBE) used to prepare and run the gel. 

Preparing and running QC reference standards at a frequency of once every 5 or 10 samples. This QC standard serves to monitor instrument precision and accuracy during a batch run. This assumes that both calibration and ICV criteria have been met. A mean value for the QC reference standard should be obtained over all QC standards run in the batch. Polyacrylamide gel electrophoresis. Standard 7.5% polyacrylamide gels were prepared and run in glycine-Tris buffer, pH 9.0 at a constant current of 25 mA at room temperature. Bromophenol Blue was used as a marker and Coomassie Brillian Blue was the staining reagent. 

To increase the quality of images, it is recommended that the nanocomposite be extruded or compression molded into a film in order to orient the clay plates. The film is then mounted in the epoxy such that the microtome slices are cut normal to the surface of the films. This will ensure that the maximum number of plates will be edge on in the TEM image. In a typical X-ray diffraction experiment, the sample can be prepared and run in an hour or two. 

Make a KBr disc of biuret and of your preparation and run their ir spectra. Assign the bands and compare the two spectra.
