Potency definition

Lead structure According to Valler and Green s definition a lead structure is a representative of a compound series with sufficient potential (as measured by potency, selectivity, pharmacokinetics, physicochemical properties, absence of toxicity and novelty) to progress to a full drug development program [12]. 

But in order to understand what needs to be changed, we first need to understand the current situation. In order for a pharmaceutical company to use any analytical method for certifying the properties (efficacy, potency, etc.) of their products, the analytical method has to be validated. Validation , in the parlance of the FDA, is a far cry from what we usually call validation when developing a multivariate spectroscopic method. In fact, what we call validation in spectroscopic calibration (which usually means calculating an SEP, or an SECV) is a far cry from the dictionary definition of validate , which is to make legally valid , where valid is defined as having legal efficacy or force [11],...

Some articles, for example, include an identical table of effective and lethal doses of high potency Fentanyl derivatives. The estimated safety margins are as high as 30,000. A competitive inhibition screen is often the first step in understanding the DDI potential of a NCE. The definitive assessment of inhibition is the inhibition constant (K ), which provides not only the inhibition potency but also information on the mechanism of inhibition (competitive, non-competitive). However in the hit to lead profiling environment this approach is over-complex for the question being asked, and generates far too many samples to enable rapid screening of compound series.
