Postcoital contraceptives estrogens

The varied clinical uses of estrogens include menstrual disorders, alleviation of menopause, associated symptoms (replacement therapy), osteoporosis, postmenopausal breast cancer and prostatic cancer and hypertrophy, lactation suppression, postcoital contraceptives, oral contraceptives (combined with progestins), estrogen deficiencies unrelated to the menopause, ovarian failure, and hypogenitalism. 

Postcoital contraception Oral contraceptive, norgestrel, mifepristone, conjugated estrogens... 

The postcoital contraceptives will prevent pregnancy if administered within 72 hours after unprotected intercourse. Several types are available, including conjugated estrogens, combined contraceptives, L-norgestrel, and mifepristone (RU 486), a progesterone antagonist that is described below. 

Other investigational or potential uses for mifepristone that are nnder development include the induction of labor after fetal death the indnction of labor at the end of the third trimester treatment of endometriosis, leiomyomas, breast cancer, and meningiomas and as a postcoital or luteal-phase contraceptive. A major concern about longterm nse is the possibility of unopposed estrogenic effects, but this concern conld be allayed by further development of selective PR modnlators. 

The biotransformation of the nonsteroidal, postcoital oral contraceptive, 2-methyl-3-ethyl-4-phenyl-A -cyclohexene carboxylic acid (Vll), was examined in monkeys and rabbits. In both species, only a trace of unaltered drug was found in the urine. At least eight metabolites were separated from rabbit urine, one of which, as yet unidentified, had 1.5 to 2.0 times the estrogenic potency of the parent compound. The antifertility effect, in the rat of certain -substituted, but not of unsubstituted compounds, was not blocked by prior treatment of the animals with SKF-525A.
