Polypeptide derivatives

It has also been possible to determine the x-ray structures of classic zinc finger motifs from several proteins bound to specific DNA fragments. We will here describe one such structure containing three zinc fingers from a mouse protein, Zif 268, which is expressed at an early developmental stage of the mouse. Hirudin is a polypeptide derived from the saliva of the leech Hirudo medicinalis that binds to the blood serine proteinase, thrombins, and thus blocks clot formation. 

Elastomeric polypeptides are a class of very interesting biopolymers and are characterized by mbber-like elasticity, large extensibility before rupture, reversible deformation without loss of energy, and high resilience upon stretching. Their useful properties have motivated their use in a wide variety of materials and biological applications. Here, we focus on two elastomeric proteins and the recombinant polypeptides derived thereof.

Although it has been shown that at least some of these polypeptides derive from exchange with the VLDL protein (E4, S22), a stringent chemical corroboration of these findings has not been published. Other minor polypeptides are also members of V, but their properties are still in the process of being defined (L2a).

Based on the failure of the research groups of Goodman (19-20) and Fegglon (21) to find adequate amounts of end groups derived from radioactive NaOCHs, sodium N-benzylcarbamate or dllsopropylamlne In polypeptides prepared by NCA polymerizations that were Initiated with these materials, many workers abandoned the Idelson-Blout mechanism. However, a number of other studies have shown that Initiator fragments can be found In polypeptides derived from NCA polymerizations that were Initiated by alkoxlde or carbamate salts (22-28). It seems that the Idelson-Blout mechanism should not be discounted based on the early radiotracer analyses. 

Specificity of the active caspase-7 ELISA was determined by analyzing captured polypeptides. Capture antibody is the caspase-7-specific antibody coated in the ELISA 96-well plates and is different from the anti-caspase-7 antibodies used for western blotting. Polypeptides that are bound by the capture antibody are referred to as captured. Jurkat cells were incubated with STS for 0-4 h and then with bzVKD-fmk for an additional 1 h. Cell extracts were incubated in 6-well plates coated with caspase-7 capture antibody. After washing, polypeptides captured on the plate were solubilized in SDS sample buffer and Western-blotted. Captured polypeptides were blotted with anti-caspase-7 LSU and anti-caspase-7 SSU to detect polypeptides derived from caspase-7. Captured polypeptides were blotted with HRP-streptavidin to detect the polypeptides covalently modified with the bzVKD-fmk inhibitor. Captured polypeptides covalently modified with bzVKD-fmk is the material that the ELISA quantifies.
