Phenothiazine Trimeprazine

The antihistaminic phenothiazine trimeprazine has significant antibacterial activity in vitro and in vivo, and a combination with trimethoprim is highly synergistic, as shown in vivo in Swiss white mice using Salmonella typhimurium as the challenge bacterium (200). 

Interestingly, penicillamine—a degradation product of penicillin and phenothiazines—the histamine H,—receptor antagonists, such as promethazine teoclate, methadilazine hydrochloride, trimeprazine tartrate are found to increase the cholesterol levels.

Trimeprazine Trimeprazine, 10-(3-dimethylamino-2-methylpropyl) phenothiazine (16.1.19), is synthesized by alkylating phenothiazine with l-dimethylamino-2-metyl-propylchloride [38].

Recent studies on multidrug reversal in mouse lymphoma and MDR/COLO 320 cells have shown that phenothiazine derivatives, namely perphenazine (2) and prochlorperazine dimaleate (4), effectively inhibited rhodamine efflux [171]. Other phenothiazine derivatives such as promethazine (1), ox-omemazine (20), methotrimeprazine maleate (18), triflupromazine (11), and trimeprazine (17) differently modulated intracellular rhodamine accumulation in these resistant cells. The effect of some substitution in the phenothiazine ring was studied in mouse lymphoma cells expressing P-gp [172], The 3,7,8-trihydroxy- and 7,8-dihydroxychlorpromazine derivatives were effective P-gp inhibitors, whereas 7,8-diacetoxy-, 7,8-dimethoxy-, 7-semicarbazone-, and 5-oxo-chlorpromazine derivatives exerted only a moderate effect.

Aly et al. [73] proposed a rapid and sensitive flow-injection chemilu-minometric method for the determination of three phenothiazine derivatives fluphenazine hydrochloride, levomepromazine hydrochloride and trimeprazine tartrate. The method was based on the chemiluminescence (CL) induced by the oxidation of the drugs with cerium (Ce(IV)) in an acidic medium. The CL intensity was greatly enhanced when rhodamine B was used as a sensitizer in the case of levomepromazine hydrochloride and trimeprazine tartrate. Trimeprazine, a phenothiazine antihistaminic possessing antipruritic activity (25 mg p.o. q.i.d.), is used in pruritis.
