The procedure for epitope mapping by the phage display method involves biopanning, amplification, assay for positive colonies, DNA isolation, and sequencing. Some steps of the procedure must be performed at the same time. Therefore, we describe the protocols in Subheadings 3.1. and 3.2. on a day-by-day manner. All steps of biopanning and phage amplification should be carried out under sterile conditions, when possible. [Pg.136]

In addition to selecting peptides for metal substrates, phage display methods successfully identified peptides capable of forming sihea nanospheres. Recently, many groups have isolated proteins and peptides responsible for the in vivo prodnetion of sihea. Stone and coworkers have isolated additional novel peptides nsing phage display that are capable... [Pg.5365]

Selection and screening methods are effective tools for shidying macromolecular interactions. Valuable methods are the yeast-based one-hybrid and two-hybrid systems (for smdy-ing protein-DNA and protein-protein interactions, respectively) and bacterial-based phage display methods (for studying either type of interaction). These systems have been used to... [Pg.22]

G. P. Smith described in a seminal paper the use of filamentous phage for the synthesis of peptide libraries (phage display method. Science, 1985,228, 1315)... [Pg.6]

Libraries synthesised using a molecular biology approach on the surface of filamentous phage particles or plasmids. These phage display methods have found wide use in protein epitope mapping approaches of proteins and protein domains. Several reviews appeared in the literature summarising the work in this field. - ... [Pg.186]

Peptides that can bind to PPV by the phage display method [161] have been identified. Aromatic amino acids were enriched in these peptide sequences, suggesting that a TT-TT interaction is the key interaction between the peptides and PPV. [Pg.91]
