Acetylation is one of the most frequently used derivatization techniques in the mass spectral analysis of peptides. Reaction with freshly distilled acetic anhydride in methanol [27] results in acetylation of a-amino functionalities within one minute. Acetylation at other less reactive sites may be accelerated by the addition of a small amount of base, such as NaHC03 or triethylamine [28], Peptide hydroxyl and amino groups can be acety-lated by exposure to a 1 1 mixture of acetic anhydride/ pyridine for about 40 minutes. The net result of the reaction is a positive shift of 42 daltons for each acetyl group addition. The failure of an acetylation reaction in a peptide analysis may be diagnostic of a blocked N-terminus. [Pg.273]

The chloroisocyanurates can be used in the bleaching of cotton, synthetics, and their blends they do, however, attack proteinaceous fibers, such as silk or wool, presumably via active chlorine reaction with the peptide (amide) linkage. However, the chloroisocyanurates can be used as shrink-proofing agents in wool finishing (131), (see Textiles Wool). The same action of chlorine upon proteins contributes to the effectiveness of chloroisocyanurates in automatic dishwashers. [Pg.422]

It is known that not all reactions proceed in the same manner on all adsorbent layers because the material in the layer may promote or retard the reaction. Because the amount of time required for a given amino acid to elute from a standard column is reproducible, the identities of the amino acids in a peptide can be determined. The amount of each amino acid in the sample is determined by measuring the intensity of the purple color resulting from its reaction with ninhydrin. Figure 26.3 shows the results of amino acid analysis of a standard equimolar mixture of 17 a-amino acids. A majority of these processes involve CO insertion to produce species that have ketene-like reactivity. Cyclo addition reactions presented include reaction with imines to form /1-lactams, with alkenes to form cyclobutanones, with aldehydes to form /1-lactones, and with azoarenes to form diazetidinones. Photoinduced benzannulation processes are included. Reactions involving nucleophilic attack to form esters, amino acids, peptides, allenes, acylated arenes, and aza-Cope rearrangement products are detailed. A number of photoinduced reactions of carbenes do not involve CO insertion. These include reactions with sulfur ylides and sulfilimines, cyclopropanation, 1,3-dipolar cycloadditions, and acyl migrations. [Pg.157]

The processes of both seed formation and fibril extension are dependent on temperature and on peptide concentration, with 37°C being required for establishing equilibrium within 24 h with 30 pM Pi 4o- A full description of the assay system may be found elsewhere [97,117], A 4 h reaction time is typically within the linear portion of the time course. This nucleus-dependent assay detects mainly inhibitors that are substoichiometric with the monomeric peptide, which is present at high concentration. It is relatively insensitive to inhibitors that target the monomeric peptide. Whether the inhibitors interact with the growing end of a seed or with a low abundance conformational form of the p peptide that is competent to add to the seed is difficult to determine at this time. Similar dose-response curves are obtained for Congo Red as an inhibitor with either thioflavin T (ThT) fluorescence or filtration of radioiodinated peptide readouts (Fig. 4) Caveats in the interpretation of both the ThT and radiometric filtration assays for the evaluation of putative inhibitors are discussed elsewhere [97]. [Pg.263]
