Pentafluoropropionic anhydride

Compound 87 in acid-hydrolyzed urine serves as a tracer for occupational exposure to the corresponding diisocyanate. It was derivatized with pentafluoropropionic anhydride and determined by LC using TSP-MS and plasmaspray (PSP) MS (discharge-assisted TSP-MS). The [M — 2](—) ion was measured instrumental LOD 0.1 pg/pL LOD about 0.2 -ig/L urine, RSD 10% for 0.5 pg/L230. Another determination of 87 in urine is with isobutyl chloroformate231.

Propranolol and related substances can be converted into TFA derivatives [558,559], which permit concentrations down to 10 ng/ml in plasma and similar samples to be analysed with the use of an ECD. The application of pentafluoropropionic anhydride [560] leads to a reduction in the time necessary for the analysis (acylation for 1 min at 70°C) and enables higher sensitivity of the determination to be obtained. 

The extracts obtained by supercritical phase extraction are evaporated vmder a current of nitrogen. Reduction of cocaine and/or benzoylecgonine with LiAlH to 2-hydroxymethyl tropine and subsequent O-acylation with heptafluorobutyric anhydride or pentafluoropropionic anhydride. 

To 5.0 ml of an aqueous solution containing cocaine is added 1 ml of saturated sodium tetraborate solution and 2.0 ml of cyclohexane. Edlund described a method for simultaneous determination of morphine, 6-0-acetyl morphine and codeine in human plasma or blood. The samples were buffered to pH 9 and extracted on silica columns, cleaned by extraction and finally acylated with pentafluoropropionic anhydride. The derivatives formed were separated on a glass capillary column (25 m by 0.36 mm I.D., coated with OV-1) at 220°C. The falling needle injection and electron capture detection were used. Although degradation of the solutes was observed, the degradation observed was very reproducible, so that quantitative analyses could be carried out in spite of the degradation. The author recommended regular control and calibration in order to obtain reliable results. 

Morphine (1 ng-1 yg) is solved in 50 yl dry ethyl acetate and 100 yl pentafluoropropionic anhydride is added. The tightly stoppered tube is placed in an oven at 60°C for 30 minutes. Excess of reagent is removed by a gentle stream of dry nitrogen and the residue dissolved in 50 yl dry ethyl acetate. Of this solution 1-2 yl is injected for oas chromatonraphic analysis19. 

Other workers [64] utilized the NICI mode for studying melatonin (iV-acetyl methoxy tryptamine) derivatized with pentafluoropropionic anhydride and showed that this method had a great sensitivity.
