Esomeprazole is the S-isomer of omeprazole (Prilosec), which is a mixture of the S- and R-isomers. Prilosec, for many years a best selling drug, is 5-methoxy-2-[[(4-methoxy-3,5-dimethyl-2-pyridinyl)methyl]sulfinyl]-IH-benzimidazole. Its empirical formula is (Ci7Hi8N303S)2Mg-3H20 with a molecular weight of 767.2 as a trihydrate and 713.1 on an anhydrous basis. [Pg.8]

Omeprazole (Prilosec) was the original PPI this drug is now joined by esomeprazole (Nexium), lansoprazole (Prevacid), pantoprazole (Protonix), and rabeprazole (AcipHex) (see Table 27-2). All of these drugs are similar, with selection often depending on cost, availability, and the drug interaction potential of each agent.15 Likewise, nonprescription forms of certain PPIs are now available, and these forms offer a convenient,... [Pg.391]

Initial results yielded drugs that were found to suppress acid secretion but were too unstable, too toxic, or insufficiently selective. However, application of the drug development process described above eventually led to better molecules and omeprazole (Prilosec) was discovered in 1978. Human testing began in 1983 and 1984 (Lind et al, 1983 and Lambers et al, 1984) and marketing approval... [Pg.101]

Similar to the evolution of omeprazole (Prilosec) to esomeprazole (Nexium), the switch from racemic modafinil to enantioenriched armodafinil utilized asymmetric oxidation of sulfide.35 Although several asymmetric oxidation methods to provide enantiopure sulfoxides have been developed, the modified Kagan system [(Ti(Oi-Pr)4/(5,5)-DET] was selected due to superior yields and optical purities (% ee).36b The Kagan method is very useful, but it is substrate dependant (Table 3). Several sulfide derivatives of modafinil were screened to determine a direction for optimization. It was found the sulfide amide 11 provided excellent optical purity and further optimization. [Pg.300]

Ranitidine (Zantac /GlaxoSmithKline) is a histamine H2-receptor antagonist that inhibits the release of gastric acid and is useful in the treatment of a variety of hypersecretory conditions [dyspepsia, heartburn, duodenal and gastric ulcers, and gastroesophageal reflux (GERD)]. Lansoprazole (Prevacid /TAP), omeprazole (Prilosec /AstraZeneca), and esomeprazole (Nexium /AstraZeneca) are benzimidazole... [Pg.415]

The current therapy leaders are the anti-ulcerants such as Omeprazole (Prilosec ) or Lansoprazole (Prevacid ), which accounted for US 19.5 billion in 2001, thus claiming a market share of 6%. The anti-ulcerants are closely followed by the cholesterol reducers, e.g., Atorvastatin (lipitor ) and Simvastatin (Zocor ), totaling US 18.9 billion, the antidepressants (US 15.9 billion), and the NSAIDs (nonsteroidal anti-inflammatory drugs US 10.9 billion). Calcium antagonists, antipsychotics, oral antidiabetics, ACE inhibitors, cephalosporins, and antihistamines complete the list of the best-selling therapy classes, with sales between US 9.9 and 6.7 billion. It is interesting to note that four classes grew more than 20% in 2001, namely the cholesterol reducers (22%), the antipsychotics (30%), the oral antidiabetics (30%), and the antihistamines (22%) [4],... [Pg.9]

Omeprazole (Prilosec, AstraZeneca) is a drug termed as proton pump inhibitor. It turns off the secretions of acid into the stomach. When less acid is produced, there is a reduced amount of acid that can flow back up from the stomach into the esophagus to cause reflux symptoms. [Pg.9]
